Our aim in this study was to determine whether berberine could inhibit HSC proliferation in vitro and prevent experimental liver fibrosis in vivo. Activated rat hepatic stellate cells (CFSCs) were incubated with various concentrations (0-20 mu g/ml) of berberine. After 48 h incubation, berberine significantly inhibited CFSC proliferation and induced cell cycle arrest in G1 phase. Real-time and Western blotting revealed that both p21 and p27 expression was markedly reduced by berberine. Berberine also decreased Akt phosphorylation and FoxO1 phosphorylation, which led to FoxO1 nuclear translocation. The results revealed a serious situation: more than 80% of the fragments are <50 ha. almost half the remaining forest is <100 m from its edges, the average distance between fragments is large (1440 m), and nature reserves protect only 9% of the remaining forest and 1% of the original forest. On the other hand, our estimates of existing Atlantic Forest cover were Galunisertib chemical structure higher than previous ones (7-8%), ranging from 11.4% to 16%.
