The PCR reaction solution (25 μl) consisted of: 0.2 μg of genomic DNA, 0.4 μM of each primer, 1 mM dNTPs, 2 mM MgCl2, 20 mM Tris–HCl, pH 8.8, 50 mM KCl, 10 mM (NH4)2SO4, 0.1% Triton X-100 and 2U Pwo DNA polymerase (Blirt SA DNA-Gdańsk, Poland). 35 cycles were performed, using the Veriti® 96 Well Thermal Cycler (Applied Biosystems, USA), with a temperature profile of 1 min MCC950 purchase at 94°C, 1 min at 60°C and 1 min at 72°C. The E. coli TOP10 cells were transformed with the ligation mixtures and transformants were examined for the presence of the ssb-like genes, using a gel retardation assay and restriction analysis.
